Potential anti-inflammatory activities of Ampelopsis grossedentata extract

International audience. Rationale: Chronic inflammation is involved in many diseases. Nowadays, research is interested in using plants for their therapeutic benefits. We evaluate in vitro, the impact of Ampelopsis grossedentata extract (AgE), which is rich in flavonoids (dihydromyricetin) and used as medicinal tea, on inflammation.Methods: i/ The impact of AgE (50 μg/mL) was investigated on the polarization of monocytes (THP1) into M0-like or M1-like macrophages measuring cytokine gene expression. ii/ AgE (10 to 100 μg/mL) was evaluated on human blood leukocytes stimulated (PMA or LPS) to evaluate COX2 and NLRP3 expression and ROS production (DHR123 fluorescence, TecanSpark®). iii/ Protein assay (Luminex® technology) was performed to study the effect of AgE on PHA-stimulated PBMCs. Data were analyzed using one-way ANOVA followed by Dunett’s post-hoc test.Results: AgE significantly decreased the gene expression of TNF-α (RQ=0.6±0.1, p<0,01) and IL-6 (RQ=0.2±0.3, p<0,01) in M1-like macrophages along with a slight decrease for IL-8 (RQ=0.8±0.4, p=0,77) and IL-1β (RQ=0.8±0.3, p=0,55). AgE decreased ROS production for all concentrations (-33±4 %, -41.0±3,3%, -49.0±3,3% and -51.0±4,4% at 10, 25, 50, 100 μg/mL respectively, p<0.05 vs control) and the expressions of COX2 (RQ=0.5±0.3, p<0,01) and NLRP3 (RQ=0.6±0.2, p<0,05). In protein assay, AgE induced a significant decrease of IFNy, IL-17a, IL-12, IL-2 (-99%±0.66, -94%±3, -61± 20% and -80±14 % respectively, vs PHA control, p<0.05). Concerning IL-1β and IL23, a non-significant decrease was observed (-24,3±16,5% and -38,0±5,2% respectively).Conclusion: AgE, through its ability to reduce the polarization of M1 as well as the production of pro-inflammatory cytokines and ROS by leukocyte, may be a new plant to value for its potential interest in inflammatory process.
