Graphical Abstract Highlights Immunostaining and 3DISCO clearing: a powerful method for studying brain connections 3D analysis of axon guidance defects in midline mutant mice Unexpected roles for Slits and Netrin-1 in fasciculus retroflexus development In Brief Clearing techniques have recently been developed to look at mouse brains, but they are complex and expensive. Belle et al. now describe a simple procedure that combines immunolabeling, solvent-based clearing, and light-sheet fluores-cence microscopy. This technique allows large-scale screening of axon guidance defects and other developmental disor-ders in mutant mice.
