International audience. The maintenance of metabolic homeostasis relies on the balanced intake of nutrients from food. Consequently, diet composition strongly impacts whole-body physiology. Dietary formulations with strong nutrient imbalances can lead to metabolic disorders, with lipids and simple sugars playing a prominent role. This unit describes how diet formulation can be modified to generate mouse models of human metabolic pathologies, and it details methodological procedures linked to dietary manipulations, including caloric restriction and introduction of a test compound.
