MBAPs link brassinosteroid signaling and cortical microtubules

International audience. Cell surface receptors govern a large part of responses to exogenous or endogenous stimuli. In plants, receptor-like kinase families are one of the critical sensors for such purposes, but their downstream pathways are yet to be fully understood, particularly in non-transcriptional branches. Our team employs BRI1, a major brassinosteroid (BR) hormone receptor, as a representative model to understand how receptor signaling impacts downstream and is regulated at the cell surface.Recently, we have identified novel BRI1 interactors, named MBAPs (MICROTUBULE-BRI1 ASSOCIATED PROTEINs), through a yeast-two-hybrid screen. In the Co-IP assay, we showed that MBAPs interact with BRI1 in a BR-independent manner. MBAPs localize to cortical microtubules, and, to be noteworthy, MBAPs-BRI1 association was found in BiFC assay. Multiple mutants of mbaps presented hypersensitivity to BRs, suggesting MBAPs are negative regulators of BR signaling. Cell biological analysis revealed that this negative regulation was driven by suppressing microtubule reorientation through non-transcriptional mechanisms. Taken together, we suggest that MBAPs serve as direct regulators of BR-dependent microtubule reorientation in a non-transcriptional manner. Ref: Delesalle et al.
