International audience. • Direct CO2-reduction to formate is found in naturalmetabolic routes such as the Wood-Ljungdahl pathway. Thereaction is catalyzed by formate dehydrogenases (FDH),which transfer 2e– from H2, ferredoxins or NADPH to CO2.• However, for organisms growing in aerobic conditions andat ambient CO2-concentrations (400 ppm), direct CO2-reduction has remained infeasible due to severe thermodynamicand kinetic constraints on the FDH reaction.
