Postmortem change in muscle fibre autofluorescence

The postmortem metabolismkinetic of rat EDL and soleus muscles wasassessed by glycogen depletion determinationwhile autofluorescent response of differentmuscle fiber types was investigated by Deep UVsynchrotron microspectroscopy at slaughter andand 24 hours postmortem. Following a 275 nmexcitation, emission fluorescence spectra showeddiscrimination depending on postmortem time(T0 versus T24h) on both muscles at 346 and302 nm and to a lesser extent at 408 and 325 nm.Taken individually, all fiber types arediscriminated but with variable accuracy, thetype IIA showing better separation of T0comparing to T24h than other fiber types.These results highlight the relevance ofusing the autofluorescent response of musclecells to rapidly assess their state of postmortemchanges.
