National audience. Le séquençage à haut débit offre de nouvelles perspectives pour le suivi de la leucémie. Nous proposons un algorithme pouvant traiter des millions de séquences, capable de différencier les réarrangements V(D)J qui s'opèrent au sein des lymphocytes, et de les quantifier. Il est désormais possible de suivre une population clonale de lymphocytes ou de lymphoblastes au cours du temps et, en pathologie, de mesurer sa réaction au traitement. La méthode proposée est implémentée dans un logiciel open-source appelé Vidjil, et testée sur plusieurs échantillons d'un patient atteint de leucémie aiguë lymphoblastique.
